ABSTRACT

Neural tube defects are a group of severe congenital malformations, including anencephaly, spina bifida, and encephalocele, which happen when neural tube fails to achieve proper closure during early embryogenesis.Mitochondria are an important site for biological oxidation and substance metabolism, as well as an important component in maintaining homeostasis of the intracellular environment.Mitochondria are also involved in the process of oocyte maturation, fertilization and embryonic development.Normal function of mitochondria is the key to the normal development of embryos.Therefore, mitochondrial dysfunction may be one of the mechanisms of neural tube closure failure.Slc25a32, SHMT2, MTHFD2/MTHFD2L, MTHFD1L and glycine cleavage system(GCS)are a series of key molecules of the mitochondrial one-carbon metabolic pathway.This paper reviews the research progress of mitochondrial dysfunction in the pathogenesis of neural tube malformation by starting from several key molecules and combining with the two main metabolic pathways of one-carbon metabolic chain and oxidative respiratory chain.

ABSTRACT

OBJECTIVE:To study metabonomics of secondary components of Astragalus membranaceus injection on leukopenia model mice. METHODS :The mice were divided into normal group ,model group ,A. membranaceus injection group (0.004 mL/g),secondary components low-dose ,medium-dose and high-dose groups (0.004,0.008,0.016 mL/g),with 7 mice in each group. Except for normal group ,other groups were given cyclophosphamide (80 mg/kg) intraperitoneally to induce leukopenia model. After modeling ,administration groups were given relevant medicine intraperitoneally ,and normal group and model group were given constant volume of sterile water for injection intraperitoneally ,once a day ,for consecutive 7 days. During the experiment ,the changes of body weight of mice were measured every 2 days. After the last administration ,the blood routine indexes [white blood cell (WBC),neutrophil(NE),lymphocyte(LY)and monocyte (MO)counts] of mice were detected by animal blood analyzer. UPLC-Q Exactive Orbitrap-HRMS combined with multivariate statistical analysis were used to analyze the metabonomics of mice serum. RESULTS :Compared with model group ,body weight of mice in the secondary component low-dose group increased significantly on the 4th and 8th day of administration (P<0.05),and the counts of WBC ,NE,LY and MO in serum of mice in secondary component groups increased significantly (P<0.05 or P<0.01). Metabonomic analysis showed that the secondary components of A. membranaceus injection could significantly regulate the contents of 8 endogenous metabolites in serum, including spermidine , uric acid , citric acid , nicotinamide, eicosapentaenoic acid , linoleic acid , erucamide and sphingosine-1-phosphate. Their effects involved linoleic acid metabolism pathway ,nicotinic acid and nicotinamide metabolism pathway and tricarboxylic acid cycle pathway. CONCLUSIONS :The secondary components of A. membranaceus injection possess the effect of increasing white blood cells in leukopenia model mice ,the mechanism of which may be related to intervention of linoleic acid metabolism pathway , nicotinic acid and nicotinamide metabolism pathway ,and tricarboxylic acid cycle pathway.

ABSTRACT

@#Conformational restriction has become one of the important strategies for modification and optimization of lead compounds in structure-based drug design. In this review, we select some recent practical examples from literature to present applications of conformational restriction in structure-based drug design. By introducing fused rings, macrocycles, spiral rings, cyclopropyl groups, bridging rings, methyl groups into molecules, we can find that the conformational restriction strategy is widely used in improving pharmacokinetic properties, enhancing the activity and selectivity, and increasing the novelty of compounds. By using these conformational restriction strategies, the drug-like properties of lead compounds can be significantly improved. This paper can help to provide theoretical guidance and practical experience for innovation in drug design and development.

ABSTRACT

OBJECTIVE:To study the intervention effects of Huan gqi injecti on on leucopenia model mice. METHODS : Kunming mice were randomly divided into normal group ,model group and drug group ,with 8 mice in each group. Model group and drug group were given intraperitoneal injection of cyclophosphamide to induce leukopenia model. Normal group was given intraperitoneal injection of equal volume of sterile water. After modeling successfully ,normal group and model group were given intraperitoneal injection of equal volume of sterile water ;drug group was given intraperitoneal injection of Huangqi injection 0.04 mL/10 g,once a day ,for consecutive 7 d. Based on the detection of blood routine indexes (leukocyte,lymphocyte,neutrophil, monocyte count )of mice in each group ,the metabolites in serum were analyzed by LC-MS. Principal component analysis (PCA), partial least squares discriminant analysis (PLS-DA),orthogonal partial least squares-discriminant analysis (OPLS-DA),HMDB, METLIN, KEGG and other databases as well as related literatures were used to identify the differential metabolites. The metabolic pathways of differential metabolites were analyzed with MetPA online tools ,and the correlation of blood routine indexes and differential metabolites were analyzed on the basis of Pearson correlation coefficient. RESULTS: Compared with normal group , blood rountine indexes of model group were decreased significantly (P< 0.01). Compared with model group ,above blood r ountine indexes of drug group were all increased siginificantly (P<0.01). LC-MS chromatogram of serum samples in normal group and model group were significantly different ,and LC-MS metabonomics data of serum samples in drug group were similar to those of normal group. Multivariate statistical analysis and correlated database analysis revealed that compared with normal group ,serum contents of 17 metabolites as L-isoleucine,eicosapentaenoic acid were increased significantly in model group ,while the contents of 4 metabolites as spermidine were decreased significantly (P<0.05 or P<0.01). Compared with model group ,Huangqi injection could reverse the serum contents of 9 metabolites in mice ,such as citric acid ,L-proline,acetylcarnitine,L-isoleucine, L-phenylalanine,sphingosine-1-phosphate,lysophosphatidylinositol,eicosapentaenoic acid and linoleic acid (P<0.05 or P< 0.01),which were associated with linoleic acid metabolism ,biosynthesis of phenylalanine ,tyrosine and tryptophan ,and phenylalanine metabolism (metabolism pathway influential values were all higher than 0.1). Correlation analysis showed that there was a significant correlation between blood routine indexes and the contents of D-sphingosine,linoleic acid and citric acid in model group(the absolute values of r were generally greater than 0.5). CONCLUSIONS :Huangqi injection can increase the counts of leukocytes,lymphocytes,neutrophils and monocytes in leucopenia model mice. The increase of leukocytes may be related to linoleic acid metabolism ,biosynthesis of phenylalanine ,tyrosine and tryptophan ,and phenylalanine metabolism.

ABSTRACT

Objective The purpose of this study was to examine the predictive value of elevated cardiac troponin I (cTnI) and Score for the Targeting of Atrial Fibrillation (STAF) in the diagnosis of cardiogenic cerebral infarction. Methods Two hundred twenty-three patients with acute ischemic stroke were recruited in the study including 38 patients in cardiogenic cerebral infarction (CCI) group and 185 in non-cardiac cerebral infarction (NCCI) group. Clinical data were collected. Chemiluminescence immunoassay was used to detect serum cTnI concentrations in patients and STAF scores were calculated. The clinical baseline data of the two groups were compared. A receiver operating characteristic (ROC) curve was used to determine the boundary value of cTnI and STAF scores in diagnosing CCI and in analyzing their predictive value. Results In the CCI group, the patients were older with higher frequency in atrial fibrillation and ischemic heart disease. Moreover, the NIHSS scores, the value of cTnI and STAF scores were significantly higher in CCI group than in the NCCI group (P<0.05). The area under the ROC curve of STAF scores was 0.954, and its 95%CI was between 0.924 and 0.985. The area under the ROC curve of the cTnI value was 0.852, and its 95% CI was between 0.788 and 0.916. The cutoff of STAF scores was 4 points, with a sensitivity of 92.1% and a specificity of 89.2%. The cutoff of cTnI value was 0.0085ng/ml, with a sensitivity of 73.7% and a specificity of 84.9%. Conclusion Serum cTnI value and STAF score have a good predictive value for CCI, and STAF score have a higher value than serum cTnI in predicting the diagnosis of CCI. Clinically, serum cTnI and STAF score may be helpful for etiology classification of acute ischemic stroke.

ABSTRACT

Objective To investigate the short-term efficacy,safety and factors affecting the efficacy of BCG intravesical therapy in high risk non-muscle-invasive bladder cancer (NMIBC) patients.Methods A total of 161 high-risk non-muscle invasive bladder cancer (NMIBC) patients were reviewed in our hospital from March 2014 to December 2017.They were all treated with BCG instillation after transurethral resection of bladder tumor (TURBT).There were 121 males (75.2%) and 40 females (24.8%).Median age was 65 years old,including 17 cases (10.6%) <50 years old,23 cases (14.3%) within 50-59 years old,72 cases (44.7%) within 60-69 years old,49 cases ≥70 years old (30.4%).There were 112 patients (69.6%) with primary bladder cancer and 49 (30.4%) patients with recurrent bladder cancer.56 cases (34.8%) had single tumor and 105 cases (65.2%) had multiple tumors.The tumors size in 106 cases (65.8%) was less than 3 cm,and tumor size in 55 cases (34.2%) was more than 3 cm.43 patients (26.7%) suffered carcinoma in situ.10 patients (6.2%) suffered urothelial carcinoma with variant types.According to the American Joint Commission for Cancer (AJCC) version 7 TNM staging system,25 cases (15.5%) were classified into Ta stage,129 cases (80.1%) were classified into T1 stage,and 7 cases (4.3%) were classified into Tis stage.There were 8 cases (5%) with low-grade cancer and 153 cases (95%) with high-grade cancer.69 patients (42.9%) received chemo-instillation before.43 cases were directly perfused without re-TURBT and 118 cases were perfused after re-TURBT.They were all treated with BCG instillation after transurethral resection of bladder tumor (TURBT).The 120 mg BCG were dissolved into 50 ml saline for instillation and were kept for 2 hours.Induction scheme of six-weekly and three fortnightly instillations started two weeks after the initial TUR or re-TUR.Maintenance instillations were then be offered in a scheme of ten monthly instillations.During treatment,patients were offered cystoscopy and cytology every three months,while CT and chest radiographs were reviewed every 6-12 months.Recurrence status and adverse effects were recorded.Univariate and multivariate regression analyses were performed to predict risk factors for failure of BCG instillation in bladder cancer.Results A total of 161 patients were followed up.The median follow-up time was 13 months,ranging 7-22 months.The overall recurrence rate was 26.1% (42/161) and the 1-year recurrence-free survival rate was 79.0%.On univariate analysis,recurrence history,history of instillation chemotherapy application and history of re-staging transurethral resection influenced recurrence.Multivariate regression analysis showed recurrence status was an independent prognostic factor regarding recurrence-free survival.The incidence of adverse events in all 161 instillation patients was 40.4% (26/65).Grade 1,grade 2 and grade 3 adverse events accounted for 53.8% (35/65),40.0% (26/65) and 6.2% (4/65) respectively.6 cases (3.7%) reduced the dose of BGC and 1 case stop the instillation due to the intolerance of BCG.Conclusion Short-term efficiency and safety were confirmed in BCG-treated high-risk NMIBC patients.And recurrence status was an independent prognostic factor for recurrence-free survival.

ABSTRACT

Objective@#To assess feasibility and clinical effect of 3D printed prosthesis in instant rehabitation of Brown Ⅱ maxilla defect.@*Methods@#Five patients with maxillary primary tumor or malignant tumor involving maxillary bone admitted from August 2016 to April 2017 were treated.Before surgery, surgical guide template and prosthesis was designed and created with digital surgery technique. Surgery was performed under guidance of surgical template and checked with surgical navigation. Prosthesis was also placed to repair Brown Ⅱ maxilla defect instantly in surgery.Two weeks after surgery, CBCT examination was taken and compared to the pre-operative CBCT images to evaluate deviation and assess precision of surgery. One month after surgery, 3D photograph was taken to compare intercommissural width, differences of bilateral lip height and maximum distance between healthy and affected sides of lip, nasolabial angles, skewed degree of mouth corner, volume variation and superficial area variation of the lip before and after insertion of prosthesis. All data ware analyzed with SPSS 21.0 using the Mann-Whitney U test for non parametric test.@*Results@#Prosthesis was inserted during 20 minutes in all 5 included patients with good occlusion. The suture was removed two weeks after surgery.The wound healed well without infection. 5 patients had no recurrence of tumor during follow-up.Maximum deviation between actual and virtual surgery was(3.75±0.87) mm, minimum deviation was 0 mm, and average deviation was(0.35±0.08) mm.The difference of bilateral lip height was(2.66±1.52) mm and(0.46±0.49) mm before and after insertion of prosthesis(P=0.016). The skewed degree of mouth corner was(3.43±1.70)°and(1.98±1.24)°before and after insertion of prosthesis(P=0.047). The maximum distance between healthy and affected sides of lip were(5.75±3.66) mm and(2.19±1.14) mm before and after insertion of prosthesis(P=0.009). The above differences were statistically significant.No significant differences were found between nasolabial angles and intercommissural width (P=0.251, 0.251). Volume variation before and after insertion of prosthesis was(5.54±2.50) cm3and superficial area variation was (66.42±21.18) cm2.@*Conclusions@#3D printed prosthesis can instantly repair Brown Ⅱ maxilla defect, retain facial appearance and dentition and obtain good effect.

ABSTRACT

Objective Studies and researches have indicated that the methylation level of PLAGL1 differentially methylated region (DMR) was associated with some development disorder syndromes.This project is purposed to prove whether methylation levels of PLAGL1 DMR is related to the fetal and early postnatal development.Methods We performed a meta-analysis of the published data on PLAGL1 DMR methylation levels in children with developmental disorders compared with that in normal children.Results PubMed,Medline,EMBASE,WanFang databases were systematically searched to identify relevant studies.We included 7 studies in this meta-analysis,with a total of 195 cases and 438 controls concerning 6 kinds of developmental disorder syndromes.The methylation level of PLAGL1 DMR was lower in children with abnormal growth (excess growth or retarded growth) than that in normal children,with a pooled percentage mean methylation difference (95% confidence intervals) of-1.05 (-1.93,-0.17).On this basis,we analyzed the odds ratio (95% confidence intervals) of hypomethylation of PLAGL1 DMR in abnormal growth children in comparison with normal children.The combined odds ratio (95% confidence intervals) of hypomethylation in abnormal growth children is 2.18 (1.23,3.88) in comparison with normal children.Conclusion Hypomethylation of PLALG1 is actually a risk factor of suffering abnormal growth for children.

ABSTRACT

Objective@#To investigate the value of prostate health index (PHI) in the diagnosis of prostate cancer in patients with total prostate specific antigen (tPSA) <20 μg/L.@*Methods@#Totally 1 135 patients with tPSA<20 μg/L and prostate biopsy indications at Department of Urology, Fudan University Shanghai Cancer Center from March 2013 to April 2016 were enrolled in this study. They were tested for serum tPSA, free prostate specific antigen and prostate specific antigen isoform 2, from which PHI was calculated. Diagnostic efficacy of PHI and tPSA were evaluated using receiver operating characteristic (ROC) curve analysis. The detection rates of prostate cancer were calculated in different ranges of PHI. Subgroup analysis of 716 patients, who were aged 50 or above with tPSA in the range of 4 to 10 μg/L and digital rectal examination negative, was performed.@*Results@#In the biopsied objects with tPSA<20 μg/L, PHI was significantly higher in prostate cancer patients than that in non-cancer patients (48.4(37.4) vs. 26.5(16.9), U=52 674.00, P=0.000), PHI was also significantly higher in high-grade prostate cancer patients than that of low-grade prostate cancer patients (44.5(30.8) vs. 56.4(42.5), U=23 314.00, P=0.000). The area under the curve (AUC) of PHI for diagnosing prostate cancer was significantly higher than that of tPSA (0.771 vs. 0.627, P=0.000). When PHI was in the range of <27, 27 to <36, 36 to <55 and ≥55, the probability of prostate cancer was 9.4% (95%CI: 7.0% to 12.2%), 16.3% (95%CI: 12.2% to 20.8%), 31.0% (95%CI: 25.9% to 37.3%) and 66.4% (95%CI: 58.9% to 74.2%), respectively. Subgroup analysis showed that the AUC of PHI in diagnosing prostate cancer was significantly higher than that of tPSA (0.764 vs. 0.569, P=0.000). When PHI was in the range of <27, 27 to <36, 36 to <55 and ≥55, the probability of prostate cancer was 8.1% (95%CI: 5.4% to 11.3%), 14.0% (95%CI: 9.1% to 19.9%), 30.8% (95%CI: 23.6% to 38.7%) and 78.8% (95%CI: 66.7% to 88.9%), respectively.@*Conclusion@#PHI is superior to tPSA in the diagnosis of prostate cancer in Chinese men with tPSA<20 μg/L.

ABSTRACT

Objective To analyze the relationship between Arg -1 expression and the clinical pathologi-cal factors ,proliferation and prognosis value in patients with colorectal cancer .Methods The expression of Arg-1 was observed in normal tissues ,chronic inflammatory tissues ,and adenomas inflammatory carcinoma tissues of mice.At the same time,Arg-1 expression was observed in human colorectal cancer adjacent tissues ,inflamed tis-sues and colorectal cancer tissues .Arg-1 expressed in 20 cases colorectal inflammation -cancer model in mice . Arg-1 expressed in 20 normal colorectal tissues .Fiftheen colitis tissues and 110 colorectal cancer tissues were examined by Immunohistochemistry .Statistical analysis was used to analyze the changes of Arg -1 expression in different groups of mice and human colon tissue cases .Results Arg-1 protein expression in normal tissues of mice was gradually increased in colon ,chronic inflammatory tissues,adenomas,inflammatory carcinoma,with sta-tistical significances(P<0.05).Arg-1 expression in para -carcinoma tissue,colitis tissues,colorectal cancer tissues was gradually increased with statistical significance (P<0.05).Conclusion Arg-1 protein is associat-ed with colorectal cancer TNM stage .Arg-1 protein may be involved in occurrence and development process of inflammation-associated colon tumor and may be a candidate of proliferated and prognostic biomarker in patients with colorectal cancer .

ABSTRACT

Objective To evaluate the effect of oral health instructions in nursing of marsupialized jaw cysts. Methods 46 patients with cysts of jaw were divided randomly into two groups (education group and control), 23 in each. In addition of normal nursing processes in both groups, patients in education group were offered more oral health instructions, such as pathogenesis of jaw cysts, mechanism of marsupialization, wear and clean of a cyst plug, and oral hygiene maintenance. 12 months later, compliance, satisfaction, treatment effect and oral hygiene condition of the patients in the two groups were studied and compared. Results 100%of patients in the education group could return to clinical visits on time and 96%of patients did cyst rinsing after every meal, which were significantly higher (χ2=6.9, P0.05). Oral hygiene condition of patients in the education group was better than control in Debris Index (χ2=9.576, P < 0.05) and Plaque Index (χ2=8.212, P < 0.05). Conclusions Oral health instructions played positive role in improving patients′ compliance, degree of satisfaction, treatment effect and oral hygiene condition in patients with jaw cysts.

ABSTRACT

@#Theranostics, combining therapy and diagnosis, is an appealing approach for chemotherapy in medicine which exhibits improved biodistribution, selective cancer targeting ability, reduced toxicity, masked drug efficacy, and minimum side effects. The role of diagnosis tools in theranostics is to collect the information of the disease state before and after specific treatment. Currently, small conjugate-based systems have received much attention for cancer treatment and diagnosis. Relative to the normal tissues, tumor microenvironment shows great difference, including acidic excessive expression of various enzymes as well as low oxygen. In this review, the latest developments on small conjugate based theranostic agents for tumor treatment and diagnosis using fluorescence were summarized.

ABSTRACT

Objective To identify allergens in portunus trituberculatus responsible for atopic dermatitis (AD) in children.Methods Totally,145 child outpatients with AD were enrolled in this study from September 2013 to July 2014,and underwent the skin prick test (SPT) with crab proteins or crab-specific IgE determination assay.Then,the children with positive SPT or elevated IgE levels underwent an oral challenge with portunus trituberculatus.Serum samples were collected from 33 children with a positive oral food challenge (test group) and from 30 health check-up child examinees (control group).Total proteins were extracted from fresh portunus trituberculatus.Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot were conducted to identify the protein fragments of portunus trituberculatus responsible for AD among these children.Results The SDS-PAGE of crude protein extracts from portunus trituberculatus yielded 11 protein bands with relative molecular masses of 94 000,70 000,58 000,49 000,36 000,34 000,32 000,27 000,21 000,19 000 and 17 000 respectively.Of the 11 protein bands,only 4 with relative molecular masses of 70 000,58 000,49 000 and 36 000 respectively reacted with sera from the patients by Western blot,with the reaction rate being 93.9%,45.4%,39.4% and 100% respectively.None of these protein bands reacted with sera from the control group by Western blot.There were significant differences between the test group and control group in the reaction rates of the four proteins with relative molecular masses of 70 000,58 000,49 000 and 36 000 respectively to sera (x2 =55.483,17.898,14.891,63.000,all P < 0.05).Conclusion The two proteins with relative molecular masses of 70 000 and 36 000 respectively are major allergens in portunus trituberculatus responsible for AD among children.

ABSTRACT

Objective To explore the role of viral infection in the development of drug eruption in patients with HIV infection, and to evaluate the efficacy of antiviral treatment. Methods This study enrolled 87 HIV-positive patients, including 11 with and 76 without drug eruption, all of whom received highly active antiretroviral therapy(HAART). Clinical data on, baseline CD4+ and CD8+ T cell counts and CD4/CD8 ratio in these subjects were retrospectively analyzed. Results The severity of drug eruption was mild in the 11 HIV-positive patients, with a mean latency period of (14.00 ± 8.10)(range, 8 - 34)days. Of the 11 patients with drug eruption, 7 had liver function impairment, which was not in accordance with the severity of skin lesions. Drug eruption was controlled in all the 11 patients after anti-anaphylactic treatment without withdrawal of antiviral drugs. Compared with 75 HIV-positive patients without drug eruption, the 11 HIV-positive patients with drug eruption showed significantly increased baseline CD4 + T cell counts (493.00 ± 245.68 (range, 42 - 810)/μl vs. 347.81 ± 167.00 (range, 11 - 814)/μl, t = 647.50, P 0.05), CD4/CD8 ratio(0.40 ± 0.27 vs. 0.29 ± 0.16, P > 0.05), or percentage of patients with a CD4/CD8 ratio below the lower limit of normal (9/10 vs. 68/69 (98.55%), P >0.05). Conclusions The latency period of drug eruption seems to be long in HIV-positive patients receiving HAART, and mild drug eruption can be complicated by liver function impairment in the patients. Relatively high CD4 + counts may be a risk factor for the development and aggravation of drug eruption in HIV-positive patients.

<p><b>OBJECTIVE</b>To identify comparatively several commercial Chinese herbal medicines and their counterfeits.</p><p><b>METHOD</b>The micromorphological characters were identified. The shape, surface, section and other characters of the medicinal materials were identified by using anatomical lens and scanning apparatus. Pictures were taken and saved.</p><p><b>RESULT</b>Main micromorphological differences between several Chinese herbal medicine including Lonicera macranthoides, L. similis, Cuminum cyminum, Plantago asiatica, Cuscuta chinensis, Sinapis alba, Salvia miltiorrhiza and their counterfeits were identified.</p><p><b>CONCLUSION</b>The reference for the authenticity identification of Chinese herbal medicine and helpful experiences for the research of the same subject were provided.</p>

ABSTRACT

ObjectiveTo explore the effects of phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt)/nuclear factor-kappa B (NF-κB) signal pathway on the process of follicle-stimulating hormone (FSH) facilitating cell proliferation and invasion in human epithelial ovarian cancer. Methods Ovarian cancer cell lines SKOV3 and 3AO were cultured to exponential phase,then assigned to control group,FSH group,LY294002 group and FSH + LY294002 group,respectively.Cells were treated with different concentration of FSH and LY294002,respectively.The effects of FSH on cell proliferation were observed by methylthiazolyl tetrazolium (MTT).Morphological changes were observed by phase contrast microscope.The ability of cell invasion was investigated by transwell invasion assay.The expression of FSH receptor (FSHR),Akt1/2,phosphorylated-Akt (p-Akt) and NF-κB p65 protein were detected by western blot.Results( 1 ) FSH could promote the proliferation of SKOV3 and 3AO cells.When the cells were treated with 40 U/L FSH for 48 hours (SKOV3) and 24 hours (3AO),compared with those in control groups,they reached the highest proliferation rate (P < 0.05 ),respectively.(2) The morphology of SKOV3 and 3AO cells in four groups:in control group,SKOV3 cells were short spindle and 3AO cells were long spindle,the nuclei of them were both roundness or oval,the cytoplasm were bright.In FSH group,the cells changed to slightly longer or polygonal,they were full in shape,meanwhile,the cell intensity were higher than control group.In LY294002 group,some cells changed from spindle to round,and began to shrink.The cell intensity diminished.The morphology of FSH + LY294002 group was similar with control group,but the cell intensity was lower than that in FSH group.(3)The number of SKOV3 cell that passed through the membrane in control group,FSH group,LY294002 group and FSH + LY294002 group was (26 ± 6),( 118 ± 19),( 18 ± 5) and ( 38 ± 7 ),respectively.The number of 3AO cell was ( 19 ± 4 ),( 134 ± 20),(12 ±3) and (58 ± 11 ),respectively.The results showed that the number of cells in FSH group was significantly higher than that in control group ( P < 0.05 ),while the number of cell in FSH + LY294002 group was significantly fewer than that in FSH group (P < 0.05 ).(4) There was no significant difference in the expression of FSHR and Akt1/2 between FSH group and control group (P > 0.05 ),but FSH increased the expression of p-Akt and the ratio of NF-κB p65 in the nucleus versus cytoplasm in SKOV3 and 3AO cells,there were significant differences compared with control group ( P < 0.05 ).LY294002 reversed the effects of FSH on increasing the expression of p-Akt and the ratio of NF-κB p65 in the nucleus versus cytoplasm,there were significant differences among LY294002 group,FSH + LY294002 group and FSH group (P < 0.05 ).ConclusionThe effects of FSH on proliferation and invasion of ovarian cancer cell lines SKOV3 and 3AO may be realized by regulating the activity of NF-κB in PI3K/Akt signal pathway.

ABSTRACT

[Objective] To investigate the therapeutic status of laparoscopic management for gallbladder stones and common bile duct stones. [Methods] A Retropective study were adopted to analysize the therapeutic scheme,effect for patients with gallbladder stones and common bile duct stones who were treated from January 1996 to September 2004. [Results] 68 cases were diagnosed, and most of them underwent single stage laparoscopic cholecystectomy (LC) and common bile duct exploration (LC BDE). A few of them were treated through endoscopic procedure in combination with LC, or open surgery when the minimal invasive surgery failed. [Conclusions] Both them indicated that patients by LC+LCBDE have shorter hospital stay, easier recovery and less complications. LC+LCBDE has obvious advantage over the other methods with appropriate operative time and high success rate. For choledochlithiasis patients, the single laparoscopic management should be a better, major option.
